Enzymatic Synthesis of Modified Nucleoside 5′-Monophosphates

There is an extensive list of applications for nucleosides, nucleotides, and their analogues that spans from substrates inhibitors in enzymatic research to anticancer antiviral drugs. Nucleoside phosphates are often obtained by chemical phosphorylation reactions, although nucleoside a promising green alternative. In this work two kinases, D. melanogaster deoxynucleoside kinase B. subtilis deoxycytidine kinase, have been employed the various canonical modified results between enzymes compared. It was determined both kinases suitable candidates 5′-monophosphate synthesis, as reaction yields 40–90% range. Deoxynucleoside however, outperforms accepts wider range substrates. Hence, were active towards 43 34 57 tested compounds, respectively. Both also larger-scale synthesis monophosphates presence GTP regeneration system using acetate E. coli.
